MBEC Reactor Protocol

BioScience Labs provides services for evaluating product efficacy in the prevention and/or removal of biofilms using in-vitro techniques. This method serves as a high throughput screening (HTS) assay to evaluate biofilm grown under batch conditions. The design allows for simultaneous testing of multiple disinfectants or one disinfectant with multiple concentrations.

Significance and Use

Vegetative biofilm bacteria are phenotypically different from suspended planktonic cells of the same genotype. Using biofilm growth reactors, biofilms are grown with specific characteristics. These characteristics can then be changed by altering the operating conditions or the engineered system.

The purpose of this test method is to use a rapid and repeatable assay to determine the susceptibility of Pseudomonas aeruginosa to multiple test materials such as antibiotics or biocides. The MBEC assay allows multiple test conditions to be run simultaneously allowing multiple concentrations of the test materials to be evaluated at once.

Scope

This test method specifies the operational parameters to grow Pseudomonas aeruginosa biofilms in a MBEC assay. The assay contains a plastic lid with 96 pegs and a plastic base (called a receiver plate) with 96 wells that can hold a total of 200 microliters. The biofilm is established on the pegs of the plastic lid with no nutrient flow in or out of the receiver plate and gentle mixing. Once the biofilms are ready, they are transferred to a new receiver plate with the test materials for disinfectant efficacy testing.

Although this test method gives the parameters to grow a Pseudomonas aeruginosa biofilm, it can be used to grow other species of biofilm.

This test method is used in the in-vitro laboratory to demonstrate disinfectant efficacy on biofilms.
