CDC Biofilm Reactor Protocol

BioScience Labs provides services for evaluation of product efficacy in the prevention and/or removal of biofilms using in-vitro techniques. This method is representative of environments where the resulting biofilm exists under high shear conditions in a liquid interface. It is used to model potable water systems, industrial water lines, oral biofilms, toilet bowls, washing machines and other scenarios where the biofilm is more firmly attached to a surface.

Significance and Use

Biofilm bacteria are phenotypically different than suspended bacteria of the same genotype. Using biofilm growth reactors, biofilms are grown with specific characteristics. These characteristics and the biofilm can then be changed by altering system parameters.

This test method will enable the growth growth of the biofilm Pseudomonas aeruginosa under high shear conditions allowing an investigator to grow, sample, and analyze the biofilm. The biofilm produced in the CDC Biofilm Reactor can also be used for efficacy testing. After the forty eight hour growth phase is done, the investigator may add a treatment in situ or remove the samples and treat them individually.

Scope

This test method specifies how to grow a Pseudomonas aeruginosa biofilm under high shear, giving a biofilm that represents high shear conditions rather than one particular environment. This method uses a CDC Biofilm Reactor that is a continuously stirred tank reactor with a high shear wall. This method also describes how to analyze a biofilm for viable cells which are recorded in colony forming units per surface area.

This test method is used in the in-vitro laboratory to demonstrate how to grow, sample, and analyze biofilms in high shear conditions.
